Glycosylation is an extreme example of a case where the modification undergoes fragmentation. Glycan profiling (also known as fingerprinting or mapping) is the best analytic method for monitoring the consistency of the glycosylation of glycoproteins. Unlike other modifications, glycosylation is the most challenging analytical project due to it is ofter a set of modifications in which a single glycosylated site on a peptide may have a number of glycan isoforms with different chain length and branches attached. It is merely possible to associated one single mass shift with glycosylation. O-glycans are linked to the protein mainly through a glycosidic bond between the terminal N-acetyl galactosamine (GalNAc) residue and the OH group of serine or threonine. o-glycan
